The first batch of all of them to be established as a primary reference standard is characterised by a variety of analytical techniques chosen to demonstrate its suitability for use (IR, UV, 1H-NMR, 13C-NMR, MS and Elemental Analysis). Full analytical testing is then performed to qualify the standard. This is the Primary Reference Standard.
Subsequent batches are established by comparison with the primary reference standard to which it is traceable. These are the Secondary Reference Standards (Working Standards).
In this study, we propose to validate the assay method used for acetone and propanol-2 (residual solvents) in a drug product, Furazidin, 50 mg, coated tablet.
The validation criteria studied were as follows:
− specificity
− limit of detection and quantification
− solution stability
− linearity
− accuracy
− precision (repeatability and intermediate precision) of the residual solvents (acetone and propanol-2) in the reconstituted pharmaceutical form and finished product.
− robustness
− analysis of several different batches of drug products.
Carry out the method for thin layer chromatography using silica gel GF254 as the coating substance and a mixture of 95 volumes of chloroform and 5 volumes of methanol as the mobile phase. Apply separately to the plate 10 µl of each of the following solutions:
A) Standard preparation
Weigh 20 mg of Furazidin reference standard in a 20 ml volumetric flask, add 15 ml of Dichloromethane, sonicate for 4-5 minutes and dilute up to the mark with Dichloromethane.
B) Test preparation
Weigh and crush 20 tablets. Weigh blend equivalent to 20 mg of Furazidin in a 20 ml volumetric flask, add 15 ml of Dichloromethane, sonicate for 4-5 minutes and dilute up to the mark with Dichloromethane. Filter the solution. After removal of the plate, allow it to dry in air and examine under ultraviolet light (254 nm).

− устойчивость
− анализ нескольких разных серий лекарственного препарата.
Проводят испытание методом тонкослойной хроматографии с использованием силикагеля GF254 в качестве вещества покрытия и смеси из 95 объема хлороформа и 5 объемов метанола в качестве подвижной фазы. Наносят на пластинку отдельно по 10 мкл каждого из следующих растворов:
A) Приготовление стандартного раствора
Взвешивают 20 мг стандартного образца Фуразидина в мерную колбу вместимостью 20 мл, добавляют 15 мл дихлорметана, обрабатывают ультразвуком в течение 4-5 минут и доводят до отметки дихлорметаном.

Взвешивают и измельчают 20 таблеток. Перед выполнением валидационных испытаний проводится проверка соответствия используемой хроматографической системы разделу фармакопейной статьи «Проверка пригодности хроматографической системы»
2. Критерии приемлемости:
время удерживания пика ацетилсалициловой кислоты на хроматограмме испытуемого раствора должно совпадать со временем удерживания пика ацетилсалициловой кислоты на хроматограмме стандартного раствора ацетилсалициловой кислоты (метод ВЭЖХ) с точностью 98,0 ‒ 102,0%
3. Около 250 мг порошка растертых таблеток АСК (используют порошок, полученный для определения примесей, раздел «Родственные соединения») помещают в стеклянный стакан или коническую колбу вместимостью 100 мл, прибавляют 50 мл воды, кипятят в течение 5 мин, охлаждают и фильтруют через мембранный фильтр с диаметром пор 0,45 мкм (RC или PET со стеклянным префильтром), отбрасывая первые порции фильтрата.
Translation - English This report reflects the results of validation of the method for the assay of amlodipine and atorvastatin in the product Statinam, 5 mg/10 mg and 10 mg/10 mg, film-coated tablets by high performance liquid chromatography (HPLC).
Before validation tests, the test on the system suitability is performed to check its compliance with the pharmacopeial monograph on the system suitability.
2. Acceptance criteria:
the retention time of the acetylsalicylic acid (ASA) peak in the chromatogram of the test solution should correspond to the retention time of the acetylsalicylic acid peak in the chromatogram of the reference solution of acetylsalicylic acid (by HPLC) with 98.0 ‒ 102.0% accuracy.
3. Place about 250 mg of powdered ASA tablets (use the powder obtained for determination of impurities, Related Compounds section) in a 100 ml glass or Erlenmeyer flask, add 50 ml of water, boil for 5 minutes, cool and filter through a 0.45-μm membrane filter (RC or PET with a glass prefilter), discarding the first ml of the filtrate.
