Source text - English Effect of diabetes on viability and mobility of murine endothelial progenitor cells.
Type 2 Diabetes Mellitus may lead to severe complications caused by micro- and macroangiopaties. They result from dysfunction of endothelial cells due to chronic hyperglycaemia. It has been shown that endothelial progenitor cells (EPC) may play a vital role in vascular biology, through induction of neovascularization in ischemic tissues or incorporation into injured vessels. Our aim was to check how thiazolidinediones, the insulin-sensitizers, affect functions of EPC isolated from healthy and diabetic mice.
Experiments were performed in the 10-week old wild type and db/db mice. Some diabetic animals were fed with rosiglitazone (10 mg/kg for 14 days), which ameliorated hyperglycaemia. EPC were purified from the bone marrow and cultured in vitro for 7-10 days. Staining of apoptotic cells with Annexin-V demonstrated that EPC isolated from the wild type mice were significantly more resistant to oxidative stress induced by H2O2 than cells isolated from db/db counterparts. Noteworthy, EPC obtained from rosiglitazone-treated diabetic individuals showed a tendency for improved viability after exposure to H2O2. On the other hand, the same treatment did not influence the increased sensitivity of EPC from db/db mice to apoptosis induced by supplementation of culture media with high concentration of glucose (25 mM). Additionally, EPCs from the untreated or rosiglitazone-treated db/db mice displayed significantly reduced migratory capabilities, which could be fully restored by rosiglitazone (10 μM) added in vitro. This effect was PPARγ-dependent, as it was reversed by supplementation of cells with PPARγ antagonist, GW9662.
Thus, rosiglitazone may improve viability and migration of EPC isolated from diabetic mice.

Сахарный диабет 2 типа может приводить к тяжелым осложнениям, вызванным микро- и макроангиопатией. Они возникают как результат дисфункции эндотелиальных клеток вследствие хронической гипергликемии. Показано, что эндотелиальные клетки-предшественники (ЭКП) могут играть ключевую роль в биологии сосудов посредством запуска реваскуляризации в ишемических тканях или встраивания в поврежденные сосуды. Наша цель состояла в том, чтобы выяснить влияние тиазолидиндионов — усилителей чувствительности рецепторов к инсулину — на функции ЭКП, изолированных из здоровых и страдающих диабетом мышей.

В то же время такая обработка не оказывала влияния на повышенную чувствительность ЭКП мышей линии db/db к апоптозу, инициированному добавлением глюкозы в высокой концентрации (25 ммоль) в питательную среду. Кроме того, ЭКП из мышей линии db/db, как получавших, так и не получавших росиглитазон, демонстрировали существенно более низкую способность к миграции, которая, однако, полностью восстанавливалась добавлением росиглитазона (10 мкмоль) in vitro. If you want to participate and sign this consent form, the study staff will do the following so the study doctor can decide if you are eligible to take part in the study:
• Ask you about your medical and surgical history, current and prior medicines, and current symptoms.
• Perform a routine complete physical examination and vital signs including taking your temperature, blood pressure, heart rate, breathing rate, and height and weight.
• Perform a 12-lead electrocardiogram, which is a painless test to check the electrical activity of the heart.
• Females only: Collect a urine sample and a blood sample for a pregnancy test, if you are a woman who is able to have children.
• Collect blood samples to determine if you are eligible to take part in the study. Your blood sample will also be tested for Hepatitis B and C. .
• Collect blood sample (8 ml - about ½ tablespoon) to check for the presence of bacteria in your blood. Blood will be drawn 2 times from 2 separate veins for a total of 4 draws.
Follow-Up Phase
Approximately one to two weeks after your last dose of the study drug, you will be asked to return to see the study doctor if the study drug worked for your infection for routine tests and check on how your infection is doing.

Source text - English TTT is a new synthetic antimicrobial agent active against Gram-positive bacteria, including methicillin resistant Staphylococcus aureus (MRSA) and vancomycin resistant Enterococcus faecium and E. faecalis. TTT binds to the 23S ribosomal RNA of the 50S subunit on the bacterial ribosome to prevent formation of an initiation complex during protein synthesis. It is efficacious in ≈ 70-75% of vancomycin resistant enterococci (VRE) infections. In Grampositive bacteria, in vitro resistance was not easily induced; serial passage of staphylococci and enterococci produced no evidence of rapid development of resistance. Nevertheless, there have now been several reports of TTT resistance emerging during treatment of enterococcal infections and a single report of resistance in a clinical strain of MRSA.
VRE have emerged as important nosocomial pathogens in the medical centers throughout the US. Over the last decade, the incidence of VRE has been increasing. In addition, these organisms are capable of prolonged survival in the hands of healthcare workers as well as on environmental surfaces commonly encountered in the healthcare setting. For these reasons, in the majority of the US hospitals and in our institution, rectal swabs for surveillance of vancomycin-resistant enterococci have been collected from patients admitted to critical departments, such as oncology, haematology and transplant units.

Reference solution (b): Dilute 1 ml of the test solution to 100 ml with a mixture of 0.5 volumes of concentrated ammonia R, 100 volumes of alcohol R and 100 volumes of methanol R. Dilute 4.0 ml to 10.0 ml with a mixture of 0.5 volumes of concentrated ammonia R, 100 volumes of alcohol R and 100 volumes of methanol R.
Plate: TLC silica gel F 254 plate R.
Mobile phase: concentrated ammonia R, methanol R, ethyl acetate R, methylene chloride R (2:40:50:90 V/V/V/V)/.
Application: 20 µl.
Development: Over 3/4 of the plate.
Drying: in air.
Detection: Examine in ultraviolet light at 254 nm.
Order of elution: ondansetron, impurity B, impurity A.
System suitability: The chromatogram obtained with reference solution (a) shows 3 clearly separated spots.
Limit:
—impurity B: any spot corresponding to impurity B in the chromatogram obtained with the test solution is not more intense than the principal spot in the chromatogram obtained with reference solution (b) (0.4 per cent).
