Most of drugs are transported as a complex with serum albumin (SA). This is an important part of drug metabolism. Bovine (BSA) and human (HSA) serum albumin tertiary structure is similar in 76% [1]. BSA is a single-chain consisting of 581 amino acids with 20 tyrosyl residues and two tryptophanes (Trp-135 in subdomain IB, Trp-214 in subdomain IIA) whereas HSA contains 585 amino acids with 17 tyrosyl residues and one tryptophan (Trp-214 in sub-domain IIA). Both serum albumins contain 17 disulfide bridges and a free thiol group (Cys 34) [2-8], They are in the shape of a heart and their dimensions are 80 x 80 x 80 x 30 A [9]. Serum albumin consists of three homologous domains (I-III), each of them is com¬posed of two subdomains (A and B) [5,9]. In the IIA and IIIA pocket the majority of small ligands, e.g. drugs are bound [3,10-12]. Binding of various endo- and exogenous ligands can results in decrease or increase of drugs affinity. Phenylbutazone (4-butyl-l,2-diphenyl-3,5 pyrazolidione, Phe) and methotrexate (4-amino-10-methylfolic acid, MTX) are commonly used to treat rheumatoid arthritis (RA). Combination of phenylbutazone and methotrexate in rheumatoid diseases is often required. Unfortunately this ther¬apy may cause competition of these drugs in binding to serum albumin and increase of uncontrolled toxic effect.

Struktura trzeciorzędowa wołowej (BSA) i ludzkiej (HSA) albuminy surowicy jest podobna w 76% [1]. Source text - English INTRODUCTION
Publications about and patents on liposomes, along with their different chemical components, preparation, and use in skincare products have often been reviewed [1-4]. The reviews do not need
any additional comments. Of interest are general questions, such as why liposomes should be used in cosmetics, what functionalities are expected from them, and what advantages they do provide
compared with alternative formulations.
The properties of the widely used main component of liposomes, phosphatidylcholine, play a key role in answering these questions. Other compounds such as niotensides and ceramides, which are
naturally predestined for the preparation of liposomes, are less important today. Niotensides do not offer superior claims, and ceramides are not available in sufficient quantities and qualities at affordable prices.
PHOSPHATIDYLCHOLINE
Looking at the horny layer, which is the barrier against external materials, phospholipids and phosphatidylcholine, in particular play, a minor role. The lipid bilayers contain only traces of
phospholipids, and the main components are free fatty acids, cholesterol, triglycerides, hydrocarbons and ceramides. But deeper in the living part of the epidermis, phosphatidylcholine is usually found as the most important constituent of all biological membranes, especially of plasma cell membranes.
Over and above that, phosphatidylcholine is the source of phosphocholine to transform ceramides into sphingomyelins. In this context, phosphatidylcholine stands for living tissues, whereas the increase of ceramides in the cells means that their death by apoptosis is imminent (Fig. 1).

Publikacje i patenty na temat liposomów, wraz z ich różnymi składnikami, przygotowaniem i wykorzystaniem w produktach pielęgnacji skóry były często omawiane [1-4]. Opinie nie wymagają żadnych dodatkowych uwag. Interesujące są ogólne pytania takie, jak dlaczego liposomy powinny być stosowane w kosmetyce, jakich funkcji się od nich oczekuje i jakie korzyści one dają w porównaniu do alternatywnych preparatów. Właściwości powszechnie używanego głównego składnika liposomów, fosfatydylocholiny, odgrywają kluczową rolę w odpowiedzi na te pytania. Pozostałe związki, takie jak niejonowe środki powierzchniowo czynne i ceramidy, które są naturalnie przewidziane do przygotowania liposomów, są dzisiaj mniej ważne. Niejonowe surfaktanty nie wykazują właściwości nadrzędnych, a ceramidy nie są dostępne w wystarczającej ilości i jakości w przystępnej cenie.
FOSFATYDYLOCHOLINA
Patrząc na warstwę rogową, która jest barierą przed zewnętrznymi materiałami, fosfolipidy i fosfatydylocholina w szczególności, odgrywają mniejszą rolę. Dwuwarstwy lipidowe zawierają tylko śladowe ilości fosfolipidów, a głównym elementami są wolne kwasy tłuszczowe, cholesterol, trójglicerydy, węglowodory i ceramidy. Ale głębiej, w części żywej naskórka, fosfatydylocholina zwykle uznawana jako najważniejszy składnik wszystkich błon biologicznych, szczególnie błon komórkowych osocza. Poza tym, fosfatydylocholina jest źródłem fosfocholiny, do przekształcenia ceramidów w sfingomieliny. W tym kontekście, fosfatydylocholina wyróżnia żywe tkanki, natomiast wzrost ceramidów w komórkach oznacza, że ich śmierć przez apoptozę jest bliska (Rys. 1).

Source text - English Summary — The Euler characteristic is applied to study the phase separation/ordering phenomena occurring in polymer mixtures. Several methods used to compute Euler characteristics are described and discussed in terms of accuracy and efficiency. A novel, robust and exact algorithm based on simplex decomposition is presented. The simulated morphology of spinodal decomposition and the phase equilibrium diagram of gradient copolymers are analyzed in terms of Euler characteristics. Again, the Euler characteristics are applied to specify the connectivity of bicontinuous patterns, to distin¬guish the disperse and the bicontinuous morphologies, and to detect the percolation transition. During the spinodal decomposition, the dynamic transformation between the bicontinuous and the droplet patterns is shown to involve the formation of a transient ''cylindrical" morphology.
Morphology is a ubiquitous feature of any multicomponent system: any spatial variation of a local property (e.g., concentration) can be treated as a complex geometrical object and viewed by drawing isosurfaces. Also, any process of phase separation or ordering can be treated as a time-dependent morphological transformation. For example, if a homogeneous binary mixture (AB) is quenched to reach the unstable part of its miscibility gap, the domains rich in component A or B will be formed shortly after the quench [1]. In the case of a symmetric mixture, a single bicontinuous interface is formed (Fig. 1a). The domains grow on average with time, while their connectivity decreases. In the case of a strongly asymmetric mixture disconnected droplets are formed at the very beginning of the phase separation process (Fig. 1b). As the average size of the domain increases, the number of droplets decreases, and shapes become more and more spherical with time. Remarkably, under some specific quench conditions, the bi-continuous can be dynamically transformed into the droplet morphology, i.e., at a certain intermediate stage of the phase separation process. These phase separation phenomena, observable in any immiscible mixture, are of extreme importance for the polymer industry, especially that the most common technological process in which two polymers are combined into a single material, is extrusion, i.e., a process of extensive mechanical mixing run at high temperatures. The strong incompatibility of different polymer species, causes that, during the extrusion process, mixing occurs only rarely on the molecular scale. After extrusion, the blend is rapidly cooled to below the glass transition or crystallization temperature. In high polymers the processes of phase separation and coalescence are very slow so that the resulting material will preserve the morphology developed on its being processed and even on further treatment involving e.g., press molding. The properties of the resulting composite will combine the properties of the raw materials in the proportion determined by blend morphology and the properties of the interface itself.
Translation - Polish Streszczenie — Zastosowano charakterystykę Eulera do badania separacji /uporządkowywania się faz, zachodzących w mieszaninach polimerów. Opisano i omówiono różne metody obliczenia charakterystyki Eulera w kategoriach dokładności i wydajności. Zaprezentowano nowy, solidny i dokładny algorytm oparty na rozkładzie sympleks. Przeanalizowano w kategorii charakterystyki Eulera zasymulowaną strukturę rozkładu spinodalnego i wykres równowagi fazowej kopolimerów gradientowych. Ponownie zastosowano charakterystykę Eulera do określenia spójności układów dwuciągłych, do odróżnienia dyspersji i morfologii dwuciągłych, oraz do wykrycia przemiany perkolacyjnej. Podczas rozkładu spinodalnego, wykazano wpływ dynamicznej przemiany pomiędzy morfologiami dwuciągłą i kropelkową, włączając tworzenie się „cylindrycznej” morfologii przejściowej.

W przypadku mieszanin symetrycznych, tworzy się pojedyncza dwuciągła granica faz (Rys. 1a). Domeny rosną średnio z czasem, podczas gdy ich spójność maleje. Te zjawiska separacji faz, obserwowane w każdej mieszaninie niemieszalnej, są niezwykle ważne dla przemysłu polimerowego, zwłaszcza że najczęstszym procesem technologicznym, w którym dwa polimery są łączone w pojedynczy materiał, jest wytłaczanie, np. proces rozległego mieszania mechanicznego prowadzonego w wysokich temperaturach. Silna niezgodność różnych gatunków polimerów powoduje, że podczas procesu wytłaczania, mieszanie rzadko zachodzi w skali cząsteczkowej. Po wytłoczeniu, mieszanka jest gwałtownie ochładzana poniżej temperatury zeszklenia lub krystalizacji. W wyższych polimerach procesy rozdziału i koalescencji są bardzo wolne, tak że otrzymany materiał zachowa morfologię rozwiniętą na jego przetworzeniu, a nawet w późniejszej obróbce, wliczając np. prasowanie. Właściwości powstałego kompozytu będą łączyć właściwości surowców, w proporcji określonej przez morfologię mieszanki i właściwości samej powierzchni międzyfazowej.

Source text - English ABSTRACT
To improve the efficiency of liposomemediated
DNA transfer as a tool for gene therapy, we have
developed a fusigenic liposome vector based on principles of
viral cell fusion. The fusion proteins of hemagglutinating virus
of Japan (HVJ; also Sendai virus) are complexed with liposomes
that encapsulate oligodeoxynucleotide or plasmid DNA.
Subsequent fusion of HVJ-liposomes with plasma membranes
introduces the DNA directly into the cytoplasm. In addition,
a DNA-binding nuclear protein is incorporated into the HVJliposome
particle to enhance plasmid transgene expression.
The fusigenic viral liposome vector has proven to be efficient
for the intracellular introduction of oligodeoxynucleotide, as
well as intact genes up to 100 kbp, both in vitro and in vivo.
Many animal tissues have been found to be suitable targets for
fusigenic viral liposome DNA transfer. In the cardiovascular
system, we have documented successful cytostatic gene therapy
in models of vascular proliferative disease using antisense
oligodeoxynucleotides against cell cycle genes, doublestranded
oligodeoxynucleotides as "decoys" to trap the transcription
factor E2F, and expression of a transgene encoding
the constitutive endothelial cell form of nitric oxide synthase.
Similar strategies are also effective for the genetic engineering
of vein grafts and for the treatment of a mouse model of
immune-mediated glomerular disease.

By ulepszyć przenoszenie DNA pośrednio przez liposom, jako narzędzie terapii genowej, opracowaliśmy fuzjogenny wektor liposomowy na podstawie reguł łączenia się (fuzji) komórek wirusowych. Białka fuzyjne japońskiego wirusa hemaglutynującego (HJV) są zespolone z liposomami, które obudowują (enkapsulują) oligodeoksynukleotyd lub plazmid DNA. Następnie fuzja HVJ liposomów z błonami plazmatycznymi wprowadza DNA prosto do cytoplazmy. Dodatkowo, jądrowe białko wiążące DNA jest włączone do cząstki HVJ-liposomu, by zwiększyć ekspresję transgenu plazmidu. Zostało udowodnione, że wektor fuzjogennego liposomu wirusowego jest efektywny dla wewnątrzkomórkowego wprowadzenia oligodeoksynukleotydu, jak również nietkniętych genów aż do 100kbp ), zarówno in vitro jak in vivo. Wiele tkanek zwierzęcych uznano jako stabilne cele dla transferu DNA fuzyjnym liposomem wirusowym. W układzie krążenia, udokumentowaliśmy pomyślną cytostatyczną terapię genową w modelach naczyniowych chorób rozrostowych, z użyciem antysensownych oligonukleotydów przeciwko genom cyklu komórkowego, dwuniciowych oligodeoksynukleotydów, jako „wabików” do złapania czynnika transkrypcyjnego E2F, i ekspresji kodowania transgenu konstytutywnej śródbłonkowej formy syntazy tlenku azotu. Podobne strategie są także efektywne dla inżynierii genetycznej przeszczepów żył i dla leczenia autoimmunologiczych chorób glomerulopatycznych na modelu myszy.

Source text - Polish Niniejsza praca ma na celu przybliżenie metody elementów skończonych ze wskazaniem na jej wady i zalety ale głownie dokonanie porównania - na prostych elementach belkowych o różnych dłógosciach, sposobie podparcia,uwierdzenia i miejsca obciążenia- różnic w obliczeniach w zależności od sposobu prowadzenia obliczeń i sposobu modelowania tych belek. Różnice te są wynikiem porównania obliczeń analitycznych oraz obliczeń dokonanych metodą elementów skończonych przy pomocy programu FEMAP przy trzech różnych sposobach modelowania: belkowym (beam), bryłowym (solid), płyowym (plate), ze wskazaniem na poprawność obliczeń danej metody. W pracy opisano ponadto schemat postępowania przy obliczeniu analitycznym belek, a takrze sposób tworzenia modelu obliczeniowego w programie FEMAP , jego sposób podparcia (utwierdzenia), obciążenia, nałożenia siatki. Wykazano w niej że przy obliczaniu belek za pomocą metody elementów skończonych przy użyciu programu Femap , najbliższe obliczeniom analitycznym są obliczenia przy użyciu elementu beam i solid natomiast element typu plate daje bardzo rozbieżne wyniki co obrazuje wykres 1 dla odkształceń i wykres 2 dla napręzeń.
Translation - English This thesis considers the finite element method, paying a particular attention to its advantages and disadvantages, but mainly compares differences in the computations, depending on the manner of calculations and modelling of simple beam elements with various length, way of supporting, fastening and place of loading. These differences are the result of comparison of analytical calculations and computations made by finite element method, using the FEMAP program with three different ways of modelling: beam, solid and plate; indicating the correctness of the given method of calculations. The thesis also describes the scheme at the analytical calculations of the beams, and also the way of creating a computational model in FEMAP program, its way of supporting, loading, and applying the mesh. It has been shown, that the calculations of beams using finite element method in FEMAP program with beam and solid elements are the most closed to the analytical calculations, while the plate type element gives very divergent results, as shown on diagram 1. for the strains, and diagram 2. for stresses.
