Selama proses pengambilan embrio, harus dipastikan media transfer selalu dalam keadaan dingin (tempatkan dalam ice bath) dalam tabung Falcon 50 ml berisi 15 ml PBS yang baru. Persiapan kultur primer embrio mencit dilakukan di Biosafety cabinet (BSC). Badan embrio dicacah halus dengan menggunakan scapel steril, lalu masukan kedalam tabung Falcon yang telah berisi PBS 5 ml dan dilakukan sentrifugasi 200 G selama 5 menit.

The mouse embryo sample is taken at the animal laboratory as the mouse embryo is put into a 50-ml Falcon tube containing 15 ml PBS with antibiotic. Throughout the embryo sampling process, the transfer media must remain cold by being placed in the ice bath. The separation of embryo organ could be conducted at the work table. Part of the embryo that is going to be cultured is only the body. Put the mouse embryo body into a new 50-ml Falcon tube containing 15-ml PBS. The preparation of mouse embryo primary culture is conducted in Biosafety Cabinet (BSC). Slice the embryo body finely with a sterile scalpel then put into a Falcon tube containing 5-ml PBS and centrifuge by 200 G for 5 minutes.
